ABSTRACT
The goal of this work is to determine the potential of the identified yeasts cultured from the palm wine samples for degradation of waste engine oil. They were selected according to their capability of degrading waste engine oil, as a carbon source in a solid medium. Samples used was fresh (Day 1), partially fermented (Day 3), and fermented (Day 7) palm wine respectively. Other analysis carried out was total colony count, cultural and morphological characteristics of the isolates. High total mean count of 18 ± 3.0cfu/ml and 58.67 ± 2.5cfu/ml in both nutrient and potato dextrose agar was recorded in fresh palm wine sample, while the lowest mean count of 5 ± 2.0 and 32 ± 3cfu/ml in both nutrient and potato dextrose agar was recorded in the fermented palm wine sample. The fungi isolated include Yeast¹, Yeast², Yeast³, Yeast⁴, Yeast⁵, Yeast⁶ Yeast⁷, Rhizopus sp.¹, Rhizopus sp.², and Rhizopus SP.³, and Mold. Organisms distribution from samples collected from the nine (9) locations sampled showed Yeast where more abundant in samples collected. While Yeasts was found in seven sources with similar characteristics (size, texture, colour), Rhizopus was identified in three sources only. The morphological characteristics of isolates showed four yeast isolates have Pseudohyphae hyphae and Ascospores spore type. The three Rhizopus isolates had Non- septate hyphae and a spore type Sporangiophore. The optical density readings of the yeast cultures at the end of the the waste oil degradation(14 days) showed Yeast⁶ recorded the highest reading of 0.45, while Yeast² and Yeast³ recorded the least value of <46. The study showed that Yeast isolates from fresh palmwine have the potential to remediate spent waste oil contaminated water.
