THE INFLUENCE OF KOLAVIRON AND MAGNESIUM ADAPTATION

ABSTRACT

Magnesium (Mg2+) supplementation has been reported to prevent cardiovascular diseases and improve endothelial function in coronary artery disease whereas Kolaviron (KV) possesses a beneficial effect in various experimental animal models through its antioxidative, and anti-inflammatory properties. The present study examined the influence of KV and Mg2+ - adaptation in acute myocardial infarction and oxidative stress in rabbits. A total of 16 adult New Zealand rabbits of both sexes with average weight of 2.3kg were obtained for this study. After 2 weeks of acclimatization, the rabbits were divided into four groups of 4 rabbits each. Group 1: (Control) were fed with normal rabbit feed +water, Group 2 were fed with high Mg2+ (4.8mM/600mg/kg bd wt.), Group 3 were fed with KV (200mg/kg bd wt.), Group 4 were fed with Mg2+ + KV (4.8mM + 200mg / kg bd wt.) for 16 weeks for KV and Mg2+ -adaption; after which two animals from each group were induced with myocardial infarction (MI) by a single dose of adrenaline (1mg/kg/ SC) on two consecutive days with an interval of 24 hours between applications and at the end of the 2nd day, oxidative stress (OS) was induced with carbon tetrachloride (CCL4) (1ml/kg/bw). Twenty-four hours thereafter, animals were anesthetized using chloroform vapour and blood sample collected via cardiac puncture for biochemical analysis.
