ABSTRACT
Methoxyacetic acid (MAA), the primary active and toxic metabolite of the industrial chemical ethylene glycol monomethyl ester (EGME), a component of paint, ink, jet fuels, varnishes. MAA causes a wide range of toxic effects including reproductive, developmental and systemic toxicity, as a result exposure to humans is inevitable. Therefore this study investigated the length of exposure effect of methoxyacetic acid on antioxidant status of the liver with histopathological changes in the liver cortex of male wistar rats. Animals were given a single dose of 450mg/kg body weight of methoxyacetic acid orally by gavage, the weight of the animals was taken at the end of each week and a certain number of the animals were sacrificed weekly. After four weeks exposure MAA had effect on gravimetric parameters, antioxidant components in the liver and plasma metabolites. Histhopathology of the liver showed severe zonal necrosis and periportal inflammatory infiltrates in the third week of analysis but the severity decreased in the fourth week of the experiment. From the experiment, although MAA-induced hepatic toxicity via single dose administration of MAA resulted in disorganization of the endogenous antioxidant system and up-regulation of inflammatory mediators in rats, the toxicological effects decreased after weeks of administration. This study has provided a new insight into the toxicity of MAA in the liver, therefore the usage of this toxicant in industries should be controlled.
