ABSTRACT
The aim of this study was to assess how different beverages impact the disintegration time of commonly used solid oral dosage formulations. Two commercially used tablet products, Aspirin and Metformin were tested. . Ten different types of beverages including orange juice, , coca cola, beer (5.15% alcohol), red wine (11% alcohol), caffeinated Bullet drink (18% alcohol), non-alcoholic malt drink and 0.1N Hydrochloric acid were compared against water. The disintegration times observed for Aspirin in distilled water was 11.33 + 1.52, Hydrochloric Acid 11.46 + 1.28, Red wine 9.66 + 0.57, in Chivita 18.67 + 2.82, in Hero Lager Beer 14.66 +1.52, Coffee 13.66 + 1.42, in Black Bullet 15.33 + 2.88, in Coca-cola 11.66 + 1.33. For Metformin in Distilled water 6.32 + 0.44, Hydrochloric acid 7.35 + 0.03, Red wine 10.88 + 0.28, Chivita 10.43 + 0.06, Hero lager beer 8.41 + 0.07, in Coffee 8.20 + 0.03, in black bullet 13.23 + 0.19, Coca-cola 5.80 + 0.04. All the beverages significantly affected disintegration times of all the tablets when compared to water.
Therefore, the tested beverages should not be used to replace water when ingesting therapeutic products. The latter part of the study was undertaken to examine the effect of the beverages etc. and their interaction on Aspirin and Metformin. Therapeutic effects in terms ofthe speed and intensity of the analgesic and anti-diabetic effect is dependent on speed of liberation from formulation. Aspirin can interact with the food or beverages, which may reduce the drug release, drug absorption in the body and may also lead to an unwanted reaction. In vitro Dissolution study was done on these beverages, using 0.1N HCL as dissolution media of pH 1.2 with USP Type II apparatus. The samples from dissolution were analyzed under U.V method.
The dissolution of drug was found to be slower in all of these beverages than in drinking water. For, Aspirin it was observed that, under the control (0.1N HCL) drug release was highest compared to other media, at 97.80% in 60 minutes. The minimum amount of drug release was observed in orange juice, 83.95% at 120 minutes. For Metformin, Orange juice also produced the highest retardation in drug release, 53.6% at 120 minutes. Thus, It is clear that these beverages influence the release profile of these medications. To get better therapeutic effects without any side effects the drug should be taken with glass of drinking water and avoid taking medicines with beverages. Medicines should not be taken with the meal to avoid any food-drug interactions. The physicians and pharmacists recognize that some foods and drugs, when taken simultaneously, can alter the body's ability to utilize a particular food or drug
