This present study focus on isolation and identification of bacteria and fungi from milk and yoghurt samples and also evaluated their phenotypic expression. Several findings suggest that milk is often contaminated by bacteria and fungi. Most of these could be pathogenic and nonpathogenic. One milliliter (1ml) each of cow milk samples were aseptically obtained from lactating cows at the cattle ranch behind Uwelu primary school in a sterile universal bottles .Samples were immediately transported in ice to the laboratory for microbiological analysis. Similarly, one brand each of peak milk and yoghurt were purchased from Uselu Market, Benin City, and taken to the lab for microbial analysis. All the analysis were carried out at 1ml each of the samples. Tenfold serial dilution were carried out by pipetting 1ml homogenate into 9ml saline water tube and labelled 10-1. A further one (1ml) was pipetted into the next tube 10. This process was repeated for all the tube to 105. One milliliter (1ml) from test tubes 103, 104were pippeted into 20ml of freshly prepared Nutrient media (NA) in a sterile agar plates in duplicates. The plates were incubated at 37o C for 24hrs (overnight) and observed colonies were counted. Bacterial isolates were further sub-cultured onto freshly prepared agar plates and incubated at 37oC overnight to obtain distinct colonies. Antibacterial susceptibility testing was carried out to determine the sensitivity of bacteria isolates to routinely used antibacterial agents like tetracycline, metronidazole, ciprofloxacin. The bacterial isolates were identified through colonial and morphological characteristic and standard and biochemical method. Fungal isolates were as well identified by visual characteristics. ,Bacterial counts obtained from human samples ranged from 6.1 x 103 to 1.8 x 104while fungal counts ranged from 9.5 x 102 to 4.6 x 103. Bacterial isolated from cow milk and yoghurt include (S. aureus, Streptococcus sp., Lactobacillus sp., Escherichia coli, Psuedomonas aeruginosa, Micrococcus sp., Bacillus sp., and Serratia marsescens). Antibacterial susceptibility testing showed that all the isolated bascterial were resistant to Gentamycin and colistin.
