ABSTRACT
Poultry are domesticated birds kept by humans for the eggs and their meat. The aim of this study is to isolation and characterizes bacterial isolate from poultry droppings. This study was carried out at the Laboratory Department of Microbiology, Faculty of Life Science, University of Benin. With the aid of sterile forceps, 1g each of poultry droppings were collected in duplicates from different poultries in three (3) different locations. Agric farm, New Benin, Ugbowo. They were put in 9ml each of sterile peptone water. Six test tubes were labeled 10-1, 10-2, 10-3, 10-4 10-5 and 10-6 with the aid of a sterile pipette, 9ml of distilled water was transferred into each test tube. The test tubes were sterilized in the autoclave and allowed to cool down. From the last three test tube (10-410-5, 10-6), 1ml was introduced into two petri dishes (in duplicate) and poured with Nutrient Agar, the petri-dishes were allowed to set and then incubated at 37oC for 24hrs in an inverted position. MacFarland standard (0.5) was prepared by mixing 0.05ml of 1% barium chloride (BaCl2) with 9.95ml of 1% Sulfuric acid(H2SO4) to barium sulphate suspension. The turbid solution (McFarland standard) formed was transferred into a test tube for comparison with bacterial inoculums suspension. Kirby-bauer disc diffusion technique was used to determine the antibacterial activity of isolated bacteria. 20ml Mueller Hinton agar plates were prepared following the manufacturer’s instructions. 1ml aliquot of each test organism suspension (standardized) was transferred onto the well-dried Mueller Hinton agar plates and was spread evenly following slow rotation of the plates and excess was decanted. The plates were allowed to dry, with the aid of sterile forcep, antibiotic disc were place in the well-dried Mueller Hinton agar plates.
