ABSTRACT
Doxorubicin is oxidized to semiquinone, an unstable metabolite, which is converted back to doxorubicin in a process that releases reactive oxygen species. Reactive oxygen species can lead to lipid peroxidation and membrane damage, DNA damage, oxidative stress, and triggers apoptotic pathways of cell death. The aim of this study was to investigate the effects aqueous extract of Cissus pulpolnae on the doxorubicin-induced testicular damage in male wistar rats Twenty (20) Adult male Wistar rats were housed in the Department of Anatomy, University of Benin. The animals were allowed to acclimatize for a period of two weeks before commencement of experiment. Cissus pulpolnae leaves were from the vegetable market at airport road in Benin City. The extract was prepared by freeze dried method at the department of medical biochemistry of the University of Benin. The rats were categorised into four groups Group A was control; group B was given 1mg/kg body weight doxorubicin intra-peritoneally only; Group C was given 500 mg/kg body weight of Cissus populnae only, Group D was given 500 mg/kg body weight of Cissus populnae and doxorubicin. At the end of the 28 days experimental period, the rats were sacrificed under chloroform anesthesia. A midline incision was made through the walls of the abdomen and the testes were dissected out and fixed in Bouin’s fluid for routine histological procedure. The result obtained showed that there was a disruption in the spematogenic series in the seminiferous epithelium of rats treated with doxorubicin. The lumen of the seminiferous tubule were devoid of spermatogenic cells. However on treatment with the extract of Cissus populnae, the spematogenic series in the seminiferous tubules was preserved and there were visible presence of spermatozoa in the lumen of the seminiferous.
