ABSTRACT
The increasing demand for continuous and quality supply of many types of mushrooms has drawn much attention and research to find potential substrate and domesticating the cultivation several species of edible mushrooms to suit local environmental conditions and resources. The selected carbohydrate grains were gotten from Uselu market, Ugbowo Benin City. The selected grains are 7 in number which are Red millet, White millet, Guniea corn, Rice, Yellow corn, White corn and Wheat. Extract of this selected grains was gotten by autoclaving 20 grams of each grains on 100 ml of water until the temperature rises to 121oC, then it was allowed to cool, then 2grams respectively of agar agar and dextrose sugar was added to some sets of the broth, while the other sets of broth was added with just 2 grams of agar agar without dextrose sugar after which the broth is poured into a petridish. Meanwhile, the culture media is ready, the Inoculum was punctured with an already sterilized Cork borer and then the punctured part of the Inoculum was collected carefully with an already sterilized Loop and is carefully inoculated at the center of the culture. Then they are being transferred to the Incubation room. From the result gotten from this study it was that found that white corn without dextrose sugar is the best substrate formulation because it supported the highest mycelia growth (6.20±0.00) at day 10 and it also influenced the biomass of the fungus growing in it. The lowest growth identified was 5.23 ± 0.04 in white millet with dextrose agar
