ABSTRACT

Liver damage, often associated with oxidative stress, is a critical health concern exacerbated by environmental toxins such as Carbon tetraachloride. The study aims to investigate the potential hepatoprotective effects of Chasmanthera dependens ethanol extract in alleviating oxidative stress parameters against CCI4 induced liver damage in Wister rats. A total of 25 male wistar rats were used for this experiment. They were grouped into 5 groups; Group 1 (normal control), Group 2 (negative control group, no treatment), Group 3 (positive control, silymarin treatment), Group 4 (ethanol extract of C.dependens treated group, 250mg/kg), Group 5 (ethanol extract of C.dependens treated group, 500mg/kg). To assess the impact, a range of oxidative stress markers were investigated, including superoxide dismutase (SOD), catalase (CAT), reduced glutathione (GSH), and malondialdehyde (MDA) levels. These markers collectively provide insights into the antioxidant defense system and oxidative damage in biological systems. The results indicate that CCI4 exposure leads to oxidative stress as reflected by a reduction in SOD, CAT, and GPx activities, along with an increase in MDA levels and a decline in GSH levels. However, treatment with the ethanol extract of Chasmanthera dependens, especially at the higher dose, demonstrates promising potential in mitigating these effects. The extract enhances antioxidant enzyme activities, reduces lipid peroxidation, and restores GSH levels, collectively suggesting a protective role against oxidative stress. These findings highlight the need for further research to uncover the precise mechanisms and therapeutic applications of this natural extract in combating oxidative stress-related ailments.
