ABSTRACT
The present study investigated the assessment of gene expressions on rat ovarian tissues treated with vitamin B complex and vitamin C during altered normal circadian clock in adult female wistar rats exposed to a model of altered photo periods. 20 female rats displaying normal estrous cycle length and weight (100g - 240g) were divided into 4 groups of 5 animals each and were administered 3.0mg/kg vitamin C and 0.2mg/kg vitamin B once a day, while the control were given normal saline and some with natural light. Temperature and weight were taken. At the end of the experiment, gene expression studies was done and the results showed that vitamin B caused a decrease in the expression of the nuclear factor erythroid 2 (Nfr-2) gene in the ovary meanwhile vitamin C caused a significant increase in the expression of the Nrf-2 gene in the ovary (p < 0.05). Vitamin B administration lowered the expression of Kelch like ECH associated protein 1 (KEAP-1). Similarly, vitamin C caused a decrease but more significantly in the expression of the Keap-1 gene in the ovarian tissue as compared with both the negative and positive control (p < 0.05). Vitamin B administration lowered the expression of Forkhead box transcription factors of the class 1 (FOXO-1) gene. Similarly, vitamin C caused a significant increase to the expression of the Keap-1 gene in the ovarian tissue as compared with the control. Significant effects were shown as p < 0.05.
