Abstract
Dumpsites are major scene in Nigeria due to poor and ineffective waste management, and could be a source of human exposure to pathogenic microorganisms. This study was aimed at investigating the prevalence of Vibrio spp. and Escherichia coli in leachate and soil from dumpsites and to detect the presence of virulence genes in them. Samples (3-lots) each were obtained from dumpsites from five different markets, all located in Edo South Senatorial District, Edo State. Bacterial isolates were isolated using standard microbiological techniques, and isolates were further identified by their morphological, biochemical and molecular techniques. The antibiotic susceptibility test and multidrug resistance profile were done by the agar disc diffusion method. DNA extraction was done by the boiling method and the presence of virulent genes (Ctx, TcpAEl tor, Zot, flicH7, Lt and Stx) in the isolates were done by polymerase chain reaction as well as gel electrophoresis. The most prevalent gene for Escherichia coli was Lt followed by FliCH7 and UidA while Stx was not detected among all isolates. A total of 31 isolates (26 Escherichia coli and 5 Vibrio spp.) had the genes (Lt, FliCH7, UidA, CtxA and TcpAEl tor) tested against whereas, no single isolate had all the genes investigated. The most prevalent gene for Vibrio spp. was CtxA followed by TcpAEl tor, whereas, Zot was not detected among all of the leachate and soil isolated. The most prevalent gene for Escherichia coli were Lt followed by FliCH7 and UidA whereas, Stx was not detected among all of the leachate and soil isolated. This study shows an increasing trend in antimicrobial resistant and virulent genes in Escherichia coli and Vibrio spp. with pathogenic potential in leachate and soil from dumpsites located in Edo South Senatorial District, Edo State.
