A STUDY ON THE PRODUCTION OF BIOSURFACTANT BY ACTINOMYCETES USING AGROWASTE

ABSTRACT

The use of cabbage leaves and yam peel as carbon source for the production of biosurfactant by actinomycetes isolated from agricultural farm land and mechanic workshop was studied. Isolation and enumeration of the actionmycetes using pour plate method revealed that the studied areas had total actinomycetes population range of 3.0 x 105cfu/g to 8.9x 105cfu/g .   Preliminary screening of all the actinomycetes isolated using submerged fermentation, drop collapse , Emulsification index (E24) and displacement methods revealed that five isolates (A1-A5) were positive to drop collapse test with the Emulsification index (E24) range of 40$ to 55.6 % and displacement values of 3.0cm to 5.0 cm.. The result of the screening for biosurfactant production of cabbage leaves and yam peel   revealed   Emulsification index values of cabbage leaves ( 40%) . Yam peel (55.6%)   and displacement values of cabbage leaves (2cm) , Yam peel 3.5 cm and the control crude oil 5cm. Determination of the biosurfactant yield using submerged fermentation and solvent extraction methods revealed that the cabbage leaves had a low biosurfactant yield of 2.3 g/l compare to the yam peel of 3.67 g/l. This study revealed that   yam peel could be a better cheap alternative source of carbon for biosurfactant production. However, a more precise quantitative and qualitative measurement of the surface tension based on availability of equipment biosurfactant   is needed.
